Combing The Role Of NMN in Improving Immunity in Three Aspects

1.4 NAD+ and SIRT1 may be the key substances to prevent the attack of new coronavirus

South African scholar R. Miller's research on NAD and SIRT1 activators as a new coronavirus treatment concluded that NAD+ deficiency can impair SIRT1 function. SIRT1 is involved in obesity-related metabolic diseases, cancer, aging, cellular senescence, cardiac aging and stress, prion-mediated neurodegeneration, inflammatory signaling responses to environmental stress, embryonic development of the brain, spinal cord and dorsal root ganglia, and placental cells play a key role in survival. SIRT1 inhibits the expression of ADAM17 against the novel coronavirus. ADAM17 is a protease-encoding gene. ADAM17 contributes to the proteolytic cleavage of cytokine receptors for TNF-α and IL-6 for their activity. The reduction of SIRT1 leads to the inability to normally suppress the expression of ADAM17, resulting in a surge of TNF-α and IL-6. COVID-19 may be caused by an uncontrolled excessive inflammatory response.

In view of the above, the activation of SIRT1 may be a key factor in the prevention of inflammatory responses, and may be necessary for successful defense against 2019-nCoV attack. Susceptible patient populations with low NAD+ levels are unable to ensure sufficient SIRT1, thus leading to failure to control 2019-nCoV replication and reduced expression of inflammatory cytokines in vivo.
