NMN, or β -Nicotinamide Mononucleotide, is a form of vitamin B3 and a key precursor of nicotinamide adenine dinucleotide (NAD⁺), which has anti-aging effects.
As a highly sought-after anti-aging star molecule in recent years, what is its safe dosage? Let's take a look at the authoritative literature reports and data.
As early as 2023, the article "Advances in Nutrition 14 (2023) 1416-1435" reviewed the safety of NMN and its clinical efficacy in humans.
This literature, integrating the results of multiple human clinical trials, presents the following views on the safety of NMN:
Existing research shows that oral NMN has good safety and tolerance within a certain dose range.
In the clinical trials that have been conducted, the dose range of NMN was 100 to 2000 mg/day, and no serious adverse events such as flushing or gastrointestinal symptoms occurred in the subjects.
There were no significant abnormal changes in vital signs (heart rate, blood pressure, blood oxygen saturation, body temperature), ophthalmic examination and sleep quality score.
The Ames test indicates that NMN is a non-mutagenic substance, further supporting its safety.
The current research has limitations, such as the majority of the subjects being healthy individuals, the sample size being relatively small (mostly 8 to 66 people), and the intervention period being relatively short (mostly 4 to 12 weeks). The safety of long-term high-dose supplementation still requires more research verification, especially the tolerance of people of different ages and health conditions needs to be further clarified.
In 2023, the research team led by Professor David Sinclair from Harvard Medical School published their findings in the journal of clinical endocrinology & metabolism. In a clinical study involving overweight or obese adults over 45 years old, Taking 2000mg of NMN daily for 28 consecutive days can not only help lose weight but also lower total cholesterol, including low-density lipoprotein cholesterol, which is known as "bad cholesterol", and also help lower blood pressure.
Taking NMN twice a day, 1000mg each time, significantly increased the NAD level in the blood from baseline to the 14th and 28th days.
The blood NAD levels in the placebo group did not change.
The NMN levels in the blood showed no significant changes, with only a brief increase 2 hours after administration on the 28th day.
For overweight or obese adults aged 45 and above, daily intake of 2000mg of NMN for 28 consecutive days showed that NMN could significantly increase the levels of NAD and its metabolites in the blood, with good safety and no serious adverse events.
It can significantly reduce body weight (by an average of 1.9kg), diastolic blood pressure (by an average of 7.01mmHg), total cholesterol (by an average of 26.89mg/dL), and low-density lipoprotein cholesterol (by an average of 18.73mg/dL), but has no significant effect on muscle function, insulin sensitivity, intrahepatic and intra-abdominal fat, etc. It provides a basis for larger-scale research.
Based on scientific data, taking 100 to 600mg of NMN per day is the ideal range for balancing safety and efficacy.
In the short term, 1000 to 2000mg per day is also safe.
The safety of long-term high-dose supplementation still requires more research for verification, especially the tolerance of people of different ages and health conditions needs to be further clarified.
