Glutathione is a low molecular weight thiol tripeptide that plays an important role in maintaining intracellular redox balance. In addition to its remarkable antioxidant properties, the discovery of its anti-melanogenic properties has led to its use as a skin lightening agent.

What does antioxidant glutathione do?
Glutathione shifts melanogenesis from eumelanin to pheomelanin synthesis through the reaction between thiol and dopaquinone, resulting in the formation of thiol-dopa conjugates. Glutathione has powerful antioxidant capacity and has been shown to scavenge reactive oxygen species produced in epidermal cells induced by UV radiation.

Glutathione can be used to treat melasma, including oral, topical and mesotherapy. The maximum dosage of glutathione reported in clinical research for whitening and freckle removal is: 500mg/day for 8 weeks or 250mg/day for 12 weeks. However, glutathione as a nutritional supplement can be safely taken orally for more than a year at a daily dose of 500mg. Glutathione can be used as an adjuvant drug, and it can be used in combination with tranexamic acid and vitamin C.

The three main routes of administration of glutathione for whitening are topical (topical lotion), oral (capsules, liposomes, and sublingual/buccal tablets) and injection (intravenous and topical).
